金雀异黄素对胃癌细胞SGC-7901凋亡的诱导及相关基因表达的调控 被引量:7

摘要 目的:研究金雀异黄素(Genistein,Gen)对胃癌细胞SGC-7901凋亡的影响及凋亡相关基因表达的调控。方法:应用MTT比色法检测Gen对胃癌细胞SGC-7901生长活力的影响;流式细胞术检测细胞凋亡;RT-PCR法检测凋亡相关基因Bcl-2、Fas表达水平的改变。结果:Gen对SGC-7901细胞有生长抑制作用,且呈时间/浓度依赖性;10.0 mg·L^(-1),20.0 mg·L^(-1)Gen能诱导SGC-7901细胞凋亡,凋亡率与剂量正相关(r=0.983 0);Gen使SGC-7901细胞Bcl-2 mRNA表达下调,Fas mRNA表达上调。结论:Gen可能通过调控Bcl-2、Fas的基因水平而诱导胃癌细胞SGC-7901凋亡。 Objective: To investigate Whether Genistein(Gen) can induce apoptosis of SGC-7901 gastric cancer cells and elucidate the regulation of Gen on Bcl-2 family genes. Method: The anti-proliferative effect of Gen was measured by MTT assay. Apoptosis was determined by flow cytometry (FCM). Alteration of mRNA of Bcl-2 and Fas genes was detected by reverse transcription polymer- ase chain reaction (RT-PCR). Result: Gen could inhibit the growth of SGC-7901 gastric cancer cells in a time/dose dependent manner, 10.0 mg · L^-1 and 20.0 mg· L^-1 of genistein dose-dependently induced apoptosis. The Fas mRNA kept increased, while the Bcl- 2 gene kept decreased during Gen treatment. Conclusion: Gen could induce apoptosis of SGC-7901 gastric cancer cells, one of the major pathways that mediated the anti-tumour response of Gen in SGC-7901 gastric cancer cells was through regulation of Bcl-2 and Fas genes expression
