摘要
目的通过观察加参方对慢性充血性心衰大鼠肺脏水通道蛋白(aquaporin,AQP)的影响,探讨加参方改善慢性充血性心衰肺淤血的机制。方法采用结扎大鼠冠状动脉前降支法造成心肌梗死后形成慢性CHF模型,随机将CHF大鼠分为加参方组、模型组和假手术组共3组,每组8只。连续给药4周。运用RT-PCR方法,对模型组,假手术组和加参方组动物肺脏AQP1、AQP4、AQP5等水通道蛋白mRNA检测,并进行统计学分析。结果慢性心衰大鼠肺脏AQP1的mRNA表达较假手术组明显上调,与假手术组相比具有显著性差异,P<0.05。加参方组大鼠肺脏AQP1mRNA表达与模型组比较显著降低,P<0.05。CHF大鼠肺脏AQP4、AQP5的mRNA表达较假手术组明显下调,与假手术组相比具有显著性差异,P<0.05。加参方组大鼠肺脏AQP4mRNA表达显著上升,与模型组比较显著P<0.05。加参方组大鼠AQP5mRNA表达与模型组相比具有显著性差异,P<0.01。结论加参方对CHF大鼠肺脏AQP1、AQP4、AQP5 mRNA异常表达产生一定影响,从而有效地清除清除支气管、脉管周围以及肺泡腔内的水份,从而有效减轻了心衰时肺水肿。
Objective To observe Jiashen prescription' effect on the expression of AQP in the lung of CHF( congestive heart failure) rats and explore Jiashen prescription' effect mechanism to relieve blood stasis in the lung of CHF rat. Methods CHFafter myocardial infarction induced by ligation of anterior descending coronary method in rats was established. They were randomly divided into three groups: Jiashen prescription group, model group and sham operated group. We used the means of RT-PCR to check the mRNA of AQP1 ,AQP4,AQPS. in the lung of CHF. Results The mRNA of AQP1 in lung of CHF rats is upregulated,compared with the normal group,P 〈0. 05. The mRNA of AQP1 in lung of jiashen prescription group rats decreased. The mRNA of AQP4, AQP5 in the lung of jiashen prescription both increased , compared with the model group. P 〈 0.01. Conclusions Jiashen prescription can effectively relieve the blood stasis of the lung of CHF rats, which is related to AQPs in the lung to sonme extent.
