原发性肝细胞癌组织VEGF-C和CD44v6表达及意义 被引量:7

EXPRESSIONS AND SIGNIFICANCE OF VEGF-C AND CD44v6 IN HEPATOCELLULAR CARCINOMA

摘要 目的探讨血管内皮细胞生长因子-C(VEGF-C)和CD44v6在原发性肝细胞癌(简称肝癌)中的表达及其与肝癌部分生物学行为的关系。方法采用常规SP免疫组织化学染色法检测33例原发性肝癌组织中VEGF-C和CD44v6的表达,并以21例肝硬化组织、16例肝外伤或肝囊肿组织作为对照,分析其表达水平与肝癌部分生物学行为的关系。结果VEGF-C定位于肝癌细胞的胞质,阳性表达率为78.8%,CD44v6主要定位于肝癌细胞的胞膜及胞质,阳性表达率为72.7%。二者在肝癌组织中的阳性表达率均明显高于癌旁组织(χ2=12.157、10.280,P<0.05);在发生转移的肝癌组织中的阳性表达率明显高于无转移的肝癌组织(χ2=4.930、8.088,P<0.05);在正常肝脏组织及肝硬化组织中几乎不表达。VEGF-C和CD44v6的表达与肝癌组织学分级、肿瘤大小、是否并发肝硬化、AFP表达、有无HBV感染无关。VEGF-C和CD44v6在肝癌组织中的阳性表达呈正相关(r=0.681,P<0.01)。结论VEGF-C和CD44v6的表达与肿瘤侵袭转移的发生有关,可作为估计预后的指标。 Objective To investigate the expressions of VEGF-C and CD44v6 in hepatocellular carcinoma (HCC) and their association with biological behavior of the disease. Methods The expressions of VEGF-C and CD44v6 in liver tissue of 33 HCC patients were examined by immunohistochemical staining, and 21 patients with portal hypertension and 16 hepatic-trauma patients served as controls. Results VEGF-C was expressed in cytoplasma in cancer tissues. The positive expression rate of VEGF was 78.8%. The expression of CD44v6 was located in membranes as well as cytoplasma in HCC, expression rate being 72.7%. The expression rates of both VEGF and CD44v6 in HCC were significantly higher than that of the adjacent tissues (χ^2= 12. 157, 10. 280;P〈0.05), VEGF and CD44v6 expressions in metastasis were significantly higher than that in the non-metastasis (χ^2 = 4. 930,8. 088;P〈0.05). Hardly any expressions of VEGF-C and CD44v6 were noted in normal liver or cirrhotic tissue. The expressions of VEGF C and CD44v6 had nothing to do with tumor size and grade, AFP level, presence or absence of cirrhosis or HBV. There was a positive correlation between VEGF-C and CD44v6 in HCC (r=0. 681,P〈0.01). Conclusion The expres- sions of VEGFC and CD44v6 are correlated with tumor metastasis and invasion, which may be used as an index for estimating the prognosis of HCC.
