RCS大鼠视网膜色素变性过程中相关细胞因子表达

THE EXPRESSION OF TNF-α,ICAM-1 AND S-100 IN RCS RAT DURING PIGMENTARY DEGENERATION OF RETINA

摘要 目的观察视网膜色素变性过程中,视网膜外节层内组织细胞肿瘤坏死因子α(TNF-α)、神经组织蛋白S-100、细胞间黏附分子-1(ICAM-1)的表达情况,探讨其在视网膜色素变性过程中的作用及意义。方法出生后25d的RCS大鼠10只,雌雄不限,12h光照-12h黑暗循环条件下饲养;以同龄SD大鼠10只作正常对照。苏木精-伊红染色和免疫组化法检测视网膜外节等范围内组织细胞TNF-α、S-100、ICAM-1等细胞因子的表达情况。结果RCS大鼠视网膜组织中TNF-α阳性表达强于SD大鼠,ICAM-1和S100阳性表达弱于SD大鼠。结论上述细胞因子的变化可能与视网膜色素上皮细胞对视细胞外节盘膜的吞噬、消化功能的改变有关。 Objective To study the expression of tumor necrosis factor-α (TNF-α), intercellular adhesion molecule-1 (ICAM-1) and S-100 in the process of pigmentary degeneration, and its signitieance. Methods Ten 25-day-old Royal College of Surgeons (RCS) rats were enrolled and 10 of age matched Sprague-Dawley (SD) rats served as the control. They were ted under a 12 h light/dark cycle. The expression of the above items by histiocytes in the photoreceptor outer segments was analyzed by Haematoxylin & Eosin (HE) staining and immunohistochemical staining. Results The TNF-α expression in RCS rat retina was more than that in SD rat. The positive ICAM-1 and S-100 in RCS rat retina were less than in SD rat. Conclusion The changes of TNF-α, ICAM-1 and S-100 expression were supposed to be related with phagocytosis and digestion of outer segment disk by the retinal pigment epithelium.
