摘要
目的探讨Notch1及其配体Dll4在骨肉瘤的表达及其和血管生成的关系。方法采用免疫组化SP法检测62例骨肉瘤Notch1和Dll4蛋白的表达,并根据CD34染色结果计数微血管密度(MVD)。43例骨软骨瘤标本作为对照。结果(1)Notch1和Dll4在骨肉瘤表达的阳性率为85.5%、88.7%,骨软骨瘤表达阳性率为21.2%、23.6%,差异具有显著性(P〈0.05);且Notch1和Dll4表达存在正相关(P〈0.05,r=0.842)。(2)Notch1和Dll4蛋白的阳性表达与骨肉瘤组织分化程度、Ennecking分期和有无转移密切相关(P〈0.05)。(3)Notch1或Dll4高表达组MVD显著高于低表达组(P〈0.05)。结论Notch1、Dll4在骨肉瘤组织中的表达上调与血管生成密切相关,可能在骨肉瘤发生、发展中起重要作用。
Objective To explorer the expression of Notch1 and its ligand Dll4 in osteosarcoma and correlation with angiogenesis. Methods the expression of Notch1 and Dll4 protein was detected through immunohistochemical SP method in 62 cases of osteosarcoma, and in accordance with CD34 staining results count microvascular density. 43 cases of osteochondroma as control specimens. Results (1)The positive expression rate of Notch1 and Dll4 protein in osteosarcoma were 85.5%, 88.7%, in osteochondroma were 21.2%, 23.6%, respectively. There was significant difference between osteosarcoma and osteochondroma groups(P〈0.05). A positive correlation between the expression rate of Notch1 and Dll4 was found in osteosarcoma group (P〈0.05, r=0.842). (2)The expression of Notch1 and Dll4 protein in osteosarcoma is closely related to differentiation, staging Ennecking and metastasis (P 〈0.05). (3)The high expression of Notch1 or Dll4 group MVD was significantly higher than the low expression group (P〈0.05). Conclusions The increased expression of Notch1 and Dll4 is closely related to angiogenesis in osteosarcoma, and probably play an important role in the development of osteosarcoma.
