摘要
[目的]了解河北省张家口市马铃薯病毒病害发生情况并对马铃薯S病毒(PVS)进行鉴定。[方法]对采自河北省张家口市的46份马铃薯样品进行生物学测定和血清学及分子生物学检测。[结果]46份样品中有35份为马铃薯Y病毒(PVY)侵染,其中有6份是PVY与PVS混合侵染,其余29份为PVY单独侵染。用特异性引物扩增PVS外壳蛋白基因的部分序列,获得685 bp的目的产物,该序列与已报道的PVS核苷酸序列同源性在80%以上,编码227个氨基酸,与已报道的PVS外壳蛋白的氨基酸同源性均在90%以上。证明该样品中确实携带PVS。[结论]该研究为今后该地区马铃薯病毒病害的控制及防治提供了依据。
[ Objective] The aim was to understand the occurrence condition of the potato virus disease in Zhangjiakou City of Hebei Province and identify the potato virus S(PVS). [ Method ] 46 potato samples collected from Zhangjiakou City of Hehei Province were detected through biological determination and serological and molecular biological detection. [Result] Among the 46 potato samples,35 samples were infected by potato virus Y(PVY) ,of which 6 sam- ples were mixed infection of PVY and PVS, the rest of 29 samples were single infection of PVY.The partial sequence of coat protein gerte of,PVS was amplified by using specific primers and the target produet of 685 bp was obtained. The sequence shared above 80% nucleotide homology with the reported PVS and it encoded 227 amino acid. The amino acid homology of the sequence was more than 90% compared with that of the reported PVS coat protein, which confimned that the sample was taking PVS. [ Conclusion] The research provided the basis for the control of potato virus disease in the area in the future.
