烟草赤星病拮抗细菌Tpb55摇瓶发酵条件的筛选

Fermentation Conditions of Antagonistic strain Tpb55 of Bacillus subtilis against Tobacco brown spot

通过单因素和正交实验相结合的方法,研究了枯草芽孢杆菌(Bacillus subtilis)Tpb55液体发酵培养液和发酵条件,筛选出最佳培养液配方为:葡萄糖1%(10 g)、蛋白胨0.3%(3 g)、MgSO4 0.05%(0.5 g)、酵母膏0.15%(1.5 g)与牛肉膏0.15%(1.5 g)的混合物,最适发酵条件为:温度31℃,初始pH7.0,发酵时间72 h。

Bacillus subtilis Tpb55 was isolated from tobacco phyllosphere and had antagonistic activity against to tobacco brown spot. Cultivation medium of Tpb55 was researched by single factor and orthogonal experiments. The fermentation conditions tested included fermentation medium, pH value before sterilized, fermentation temperature and fermentation time. The results showed that the optimal cultural liquid consisted of 1% glucose （10 g）, 0.3% peptone （3 g）, 0.05% Mgso4 （0.5 g）, 0.15% yeast extract （1.5 g） and 0.15% （1.5 g） beef extract. The optimal fermentation conditions of stain Tpb55 were predicted as below： temperature 31℃, pH7.0 and 72h, agitation 180r/min.
