摘要
目的观察川芎嗪对血管再狭窄大鼠血管紧张素Ⅱ(AngⅡ)、内皮素(ET)与血小板源生长因子(PDGF-B)的影响。方法制备大鼠血管再狭窄模型,观察颈总动脉形态学变化,放射免疫法测定血浆AngⅡ与ET含量,免疫组化法观察颈总动脉PDGF-B表达。结果颈总动脉损伤后14 d,平滑肌细胞大量增殖,AngⅡ、ET含量显著升高(均P<0.01),平滑肌PDGF-B表达显著增强(P<0.01)。川芎嗪抑制平滑肌细胞增殖,显著降低AngⅡ、ET含量及PDGF-B的表达。结论川芎嗪通过降低AngⅡ、ET含量及抑制PDGF-B表达实现抗血管再狭窄作用。
Objective To observe the effects of ligustrazine on endothelin and angiotensin Ⅱ levels and expression of PDGF - B in carotid artery restenosis rats. Methods Restenosis model was established and the rats were treated with ligustrazine from three days before operation. The morphologic change and levels of Ang Ⅱ and ET were measured. The expression of PDGF - B was de- tected by immunohistochemical technology. Results Compared with sham, the sections of injured carotid artery showed large numbers of proliferating VSMCs throughout intima and media, the levels of AnglI and ET were obviously increased( P 〈 0.01, respec- tively) , the expression of PDGF- B was significantly increased(P 〈0. 01 )in operation group. VSMC proliferation was inhibited and the ET and AnglI levels, expression of PDGF - B were decreased obviously by ligustrazine. Conclusion Ligustrazine can pre- vent restenosis through reducing the AngⅡ , ET levels and inhibits the PDGF - B expression.
