摘要
目的观察芪参益气滴丸对急性心肌梗塞(AMI)大鼠不同时间点转化生长因子-β1(TGF-β1)的影响。方法制备AMI大鼠,随机分成假手术组、AMI模型组、芪参益气滴丸治疗组[3.5 mg/(100 g.d)-1芪参益气滴丸混悬液灌胃给药]、卡托普利治疗组[1.0125 mg/(100 g.d)-1卡托普利悬浊液灌胃给药],每组又分4个时点(3 d,2周,4周,8周),共16个组,每组9只。于实验结束后处死动物,分别取大鼠梗塞区及非梗塞区心肌,RT-PCR法检测TGF-β1的mRNA表达水平。结果AMI组TGF-β1mRNA表达在梗塞区和非梗塞区与假手术组比较均呈现高表达趋势(P<0.05);芪参益气滴丸治疗组与AMI组比较,梗塞区3 d,2周时TGF-β1mRNA高度表达(P<0.05);4,8周时TGF-β1mRNA表达下降,但两组无显著性差异;非梗塞区各时间点TGF-β1mRNA表达均下降(P<0.05)。卡托普利治疗组与AMI组比较,梗塞区各时间点TGF-β1mRNA表达均下降,3 d,2周时两组有显著性差异(P<0.05);非梗死区各时间点TGF-β1mRNA表达均下降(P<0.05)。芪参益气滴丸治疗组与卡托普利治疗组比较,在梗塞区各时间点TGF-β1mRNA表达均升高,3d,2周,8周时两组有显著性差异(P<0.05);在非梗死区各时间点TGF-β1mRNA表达均下降,但两组比较无统计学意义。结论芪参益气滴丸具有促进梗塞区心肌早期修复,同时抑制非梗塞区心肌胶原的增生,从而延缓心室重构的作用。
Objective To observe the effects of Qishenyiqi Dripping Pills on the expression of TGF -β1 mRNA in rats with acute myocardial infarction(AMI) at different time point. Methods Myocardial infarction model was replicated by ligation left anterior descending branch of coronary artery in Wistar rats. Rats were randomly divided into sham operation group, AMI group, Qishenyiqi Dripping Pill group, Captopril group, and each group was divided into d different time points(3 days ,2 weeks,4 weeks,8 weeks). The levels of TGF-β1 mRNA in the no infarct and infract area of left ventricle were examined by RT - PCR. Results The mRNA expression of TNF-αin AMI group was significantly elevated as compared with sham rats ( P 〈 0.05 ). TNF -β1 in Qishenyiqi Dripping Pills group and Captopril group were markedly attenuated compared with MI group and there were significant differences in 2nd week ,4th week and 8th week( P 〈 0.05 ). TNF -αin infarct area of Qishenyiqi Dripping Pills group was markedly attenua- ted as compared with Captopril group ( P 〈 0.05 ). TNF -β1 in the no infract area of Qishenyiqi Dripping Pills group was markedly attenuated compared with Captopril group or 3rd day and in 2nd week ( P 〈 0.05 ). Conclusion Qishenyiqi Dripping Pills can in- hibit inflammatory reaction after acute myocardial infarction, relieve degree of myocardial injury, so as to protect cardiac function.
