联合应用阿霉素和姜黄素增强人白血病细胞株HL-60对阿霉素的敏感性 被引量:4

摘要 目的研究姜黄素是否增强人白血病细胞株HL-60对阿霉素的敏感性及其作用机制。方法采用MTT法测定药物的体外杀伤作用,应用流式细胞术分析细胞凋亡。应用RT—PCR和Western blot检测Survivin和XIAP基因的表达。结果姜黄素(2.00~16.00 μmol.L-1)和阿霉素(0.08~0.64μmol·L-1)同时给药对HL-60细胞可产生增强的协同杀伤效果,同时,细胞凋亡增加,Survivin和XIAP的表达下降。结论联合应用阿霉素和姜黄素可以增强人白血病细胞株HL-60对阿霉素的敏感性,下调生存蛋白(Survivin)和x-相关凋亡抑制蛋白(XIAP)表达促进细胞凋亡是其主要作用机制。 Objective To investigate the effect of curcumin on the sensitivity of human leukemia cell line HL - 60 to adriamycin. Methods Cytotoxicity was determined by the colorimetric MTT viability/proliferation assay. Apoptosis was assessed by flow cy- tometry. The mRNA levels of survivin and XIAP were measured by RT - PCR. The protein levels of survivin and XIAP were measured by Western blot. Results Simultaneous administration of curcumin ( 2.00 - 16.00 μmol ·L- 1 ) and adriamycin ( 0.08 - 0.64μmol · L- 1 ) resulted in synergistic cytotoxicity and apoptosis for HL - 60 cells. Meanwhile, cell apoptosis was increased and the mRNA and protein levels of Survivin and XIAP were significantly down - regulated. Conclusion Curcumin can sensitize human leukemia cell line HL - 60 to adriamycin - mediated apoptosis. Down - regulation of Survivin and XIAP gene expression and promoting cell apoptosis are main mechanisms.
