摘要
目的探讨黄芪对糖尿病大鼠心肌组织一氧化氮(NO)含量和一氧化氮合成酶(NOS)活性的影响。方法正常大鼠、糖尿病大鼠及高、低剂量黄芪(10,8g.kg-1.d-1)治疗8周的糖尿病大鼠各10只,分别测定各组大鼠血清乳酸脱氢酶(LDH),肌酸激酶(CK)含量及心肌组织一氧化氮(NO)含量及一氧化氮合成酶(NOS)活性。结果第8周末终止实验时,糖尿病大鼠血清LDH,CK水平和心肌NO含量及NOS活性均明显高于正常组(P<0.01);黄芪治疗组血清LDH,CK和心肌NO含量及NOS活性均明显低于糖尿病组(P<0.01),但高于正常组(P<0.01或P<0.05)。黄芪高、低剂量组间在血清LDH水平、心肌NO含量上有明显差异(P<0.05)。结论黄芪可提高心肌组织NO含量和NOS活性,对糖尿病大鼠心肌具有保护作用。
Objective To investigate the effects of astragalus on NO content and NOS activity in myocardial tissue in diabetic rats. Methods Forty male wistar rats were randomly divided into four groups: normal control group (NC), diabetic group (DM), astragalus high doses group (AS2 ,10g · kg -1· d-1) and astragalus low doses group (AS1, 8g · kg -1· d-1 ). At the end of 8 weeks ,the rats were killed. Serum lactate dehydrogenase(LDH) and creatine kinase(CK) were measured . Content of nitric oxide (NO) and activity of nitric oxide synthase(NOS) in myocardial tissues were tested. Results LDH and CK levels of serum , content of NO and activity of NOS in myocardial tissues of diabetic rats were significantly higher than those of normal con- trols ( P 〈 0.01 ). After 8 weeks of treatment, LDH and CK levels of serum , content of NO and activity of NOS in myocardial tis- sues of astragalus groups were greatly lower than those of diabetic rats, but higher than normal controls ( P 〈 0.01 or P 〈 0.05 ). There was significant difference in level of LDH, content of NO between ASl and AS2. Conclusion Astragalus can decrease NO content and NOS activity in heart of diabetic rats and has protective effect on diabetic rats.
