正交设计法优选戒毒胶囊的提取工艺 被引量:4

摘要 目的优选戒毒胶囊中挥发油、醇及水提取工艺。方法采用正交设计法,以挥发油得率为指标,筛选制剂中挥发油的提取工艺;以黄芩苷的含量为考察指标筛选醇提取工艺;以芍药苷的含量为考察指标优选金银花、白芍、黄芪等药味的水提工艺。结果挥发油的最佳提取工艺为当归、川芎加水10倍量,浸泡1 h,提取8 h;醇提最佳工艺为丹参、黄芩、栀子采用70%乙醇,溶媒6倍量,回流时间为1.5h,提取次数为2次;水提取的最佳工艺为将白芍、黄芪、龟甲和熟地、黄连同上述药渣,加10倍量水,煎煮2次,1.5 h/次,金银花于第2次煎煮时加入。结论通过正交实验优选出戒毒胶囊最佳制备工艺稳定可行。 Objective To optimize the extraction process of Jiedu Capsules. Methods The series of tests were carried out with orthogonal design. The conditions were evaluated with the yield of volatile oil as the index. With the yield rate of baicalin as the in- dex, and the concentration of alcohol, solvent volume, extracting time and times as factors, the optimal extracting process for the extracting with alcohol was studied. With the yield rate of paeoniflorin as the index, and the volume of water, extracting time and times were optimized for the water extraction. Results The optimal extracting process of volatile oil was as follows : 10 volumes of water were added and immersed for I h, and then extracted for 8h. The results of alcohol extraction was defined as 85% of alco- hol, 1.5 hours of refluxing time with 10 times volume of solvent and twice extraction. The best extracting process by water was de- fined as 1.5 hours of each extraction with 10 times volume of water and twice extraction. Flos Lonicerae was extracted at the sec- ond extraction. Conclusion The optimum preparation technology of Jiedu Capsule is suitable and reproducible.
