摘要
比较应用了2种方法制备的抑郁症大鼠模型在行为学及血清皮质醇水平的异同点,为今后抑郁症发病以及治疗机制的研究奠定基础.应用慢性温和性刺激制备外源性抑郁症大鼠模型,通过在出生第8d的幼鼠腹腔中注射5-羟色胺再摄取抑制剂—–氯米帕明制备成年内源性抑郁症大鼠模型.2种模型抑郁症大鼠在强迫游泳实验中均表现出不动时间明显延长,上窜时间明显缩短(P<0.01).在开野时间中,慢性刺激组大鼠的水平跑格数和直立次数均明显减少(P<0.05),而氯米帕明组大鼠的水平跑格次数则明显增加(P<0.05),直立次数明显减少(P<0.01).2种抑郁症大鼠的血清皮质醇水平均明显增加(P<0.01).慢性刺激模型和氯米帕明模型均复制出了人类抑郁症的多项表现,说明环境因素的改变和中枢5-羟色胺水平的降低在抑郁症发病机制中具有重要的作用.
The characteristics of behavioral model tests for the rats suffering depression are compared. In order to evaluate the results of study on depression from two perspectives, the depression is induced by both Clomipramine (CLI) and chronic mild stress in forced swimming test and open field test. The adult depressed rats are prepared by exposing rats to chronic mild stress (CMS) and by chronically injecting the tricyclic antidepressant CLI in the rats during their neonatal state of life, in which two types of animal depression models are thus built accordingly. In the forced swimming test, the immobile time for the depressed rats of both types are increased, and accordingly the climbing time are decreased (P〈0.01). The crossing times and rearing times for chronic mild stressed rats are decreased in open field test (P〈0.01). The crossing times of CLI rats, in contrast, are increased (P〈0.01), and the rearing times of CLI rats are also reduced (P〈0.01). The level of serum cortisol of depressed rats of both two types are found to be heightened (P〈0.05). Many human depression disorders can be represented with both models established for rates suffering chronic mild stress and clomipramine, indicating that the changes of environment and decrease in 5-HT have some effect in the pathogenesis of depression.
