摘要 目的:建立沙枣中鞣质含量测定的方法,优选鞣质类成分的超声提取工艺。方法:采用磷钼钨酸-干酪素比色法,通过单因素分析法考察不同溶剂对沙枣总鞣质提取率的影响,采用正交试验设计,考察提取时间、提取次数和料液比3个因素时超声提取工艺的影响。结果:在760nm波长处,没食子酸的反应物在0.001~0.01mg.ml^-1浓度范围内与吸收度呈线性关系,r=0.9994。以70%丙酮为溶剂,超声提取3次,每次1.0h,料液比为1:10时,提取效果最好,干浸膏中总鞣质含量为6.32%。结论:该比色法可以作为沙枣鞣质的含量测定和质量控制的方法,优选工艺稳定,可行。 Objective:To establish a uv- vis method for determining the contents of tannin in elaeagnus angustifolia L. ,optimize the extracting process of tannins by ultrasonic wave.Methods: The Phosphomolybdium Tungsten Acid- Casein Reaction was used. The influences of different solvents to the extracting of tannin was tested by single factors, based on that the affects of 3 factors:dine for extracting, extracting times,material and solvent ratio to ultrasonic extraction technology were performed by orthogonal design.Results: The linearity of products was obtained within the range of 0.001- 0.01 mg· ml^- 1 ( r = 0. 9994)for Gallic acid at the wavelength of 760nm. The optimum extraction conditions were as follows:adding 10 - fold of 70% acetone,extracting 3 times,each time for 1 .Oh by uhrasonic wave.The average contents of total tannin in drying extract was 6.32%. Conclusion: The method established could be use for assay the contents of tannin in elaeagnus angustifolia L. and quality control, the technology optimized were steady and reproducible.
