摘要
目的研究健脾补肾方(Jian Pi Bu Shen Fang,JPBSF)对氯胺酮所致小鼠记忆障碍的影响。方法用跳台法测定小鼠训练时和24h后重测时的错误次数。分别于训练前20min腹腔注射氯胺酮和训练前10min腹腔注射氢溴酸樟柳碱,复制2种小鼠记忆获得障碍模型。用氨基酸自动分析仪测定小鼠脑内谷氨酸的含量。结果氯胺酮模型组小鼠学习记忆实验重测后错误次数明显多于对照组(P<0.001),JPBSF小剂量+氯胺酮组、JPBSF大剂量+氯胺酮组和吡拉西坦+氯胺酮组小鼠学习记忆实验重测后错误次数明显少于氯胺酮模型组(P<0.01),氯胺酮模型组和JPBSF小剂量+氯胺酮组小鼠学习记忆实验重测后脑内谷氨酸含量差异无统计学意义(P>0.05);樟柳碱模型组小鼠学习记忆实验重测后错误次数明显多于对照组(P<0.001),JPBSF小剂量+樟柳碱组、JPBSF大剂量+樟柳碱组和吡拉西坦+樟柳碱组小鼠学习记忆实验重测后错误次数明显少于樟柳碱模型组(P<0.01)。结论JPBSF对氯胺酮造成小鼠记忆获得障碍有明显的改善作用,但不增加小鼠脑内谷氨酸含量,JPBSF对樟柳碱造成小鼠记忆获得障碍亦有明显改善作用。
Objective To investigate the effect of Jiaupibushenfang(JPBSF) on Ketamine-indueed memory impairment in mice. Methods Memory errors of mice in training and retesting 24 hours later were measured by Step-down method. The memory acquisition impairments were induced by i. p Ketamine 20 and Anisodinc 10 minutes before training in Step-down performance, respectively. Glutamate in mice brain was analyzed by automated amino acid assay. Results Memory errors of saline + ketamine group in retesting was more significant than that of saline + saline group ( P 〈 0. 001 ) , while memory errors of JPBSF low dose + ketamine group, JPBSF large dose + ketamine group and piracetam + ketamine group were significantly milder than that of saline + ketamine group ( P 〈 0.01 ). There was no difference in glutamate content in mice brain between JPBSF low dose + ketamine group and saline + ketamine group(P 〉 0.05 ). Memory errors of saline + anisodine group in retesting was obviously more than that of saline + saline group ( P 〈 0. 001 ) , while memory errors of JPBSF low dose + anisodine group, JPBSF large dose + anisodinc group and piracetam + ketamine group were significantly milder than that of saline + anisodine group(P 〈 0.01 ). Conclusion JPBSF has improved ketamine and anisodine-induced memory acquisition impairment, but did not change the glutamate level in mice brain.
