结肠肿瘤组织中MMP-2和MMP-7表达的临床研究

Expression of MMP-2 and MMP-7 in colon adenoma and colon cancer

目的:观察正常结肠黏膜、结肠腺瘤和结肠癌组织中MMP-2和MMP-7的表达,探讨两种蛋白表达在结肠肿瘤发生、侵袭及转移中的意义。方法:采用RT-PCR方法检测40例结肠腺瘤、60例结肠癌肿瘤组织和周围正常黏膜组织中MMP-2、MMP-7的表达,进行半定量分析。结果:结肠腺瘤组织中的MMP-2表达水平高于正常黏膜(P<0.05),结肠癌组织中MMP-2和MMP-7的表达水平均高于结肠腺瘤和正常黏膜(P均<0.05),Ⅲ、Ⅳ期结肠癌组织MMP-2、MMP-7的表达水平高于Ⅰ、Ⅱ期(P<0.05),Ⅳ期结肠癌组织MMP-2、MMP-7的表达水平高于Ⅲ期(P<0.05)。结论:MMP-2和MMP-7的高表达可能与结肠肿瘤的侵袭和转移有关,但其高表达的机制尚需进一步探讨。MMP-7水平的检测可用于结肠癌的早期诊断。

Objective： To investigate the expression of matrix metalloproteinase- 2（MMP- 2） and matrix metalloproteinase- 7（MMP- 7）in colon adenoma and colon cancer. Methods： Forty tissue samples of colon adenoma and 60 of colon cancer were collected .Normal colonic tissues were also collected from the same subjects. The mRNA expression levels of MMP- 2 and MMP- 7 were quantitated using semiquantitative reverse transcription polymerase chain reactions. Results： The expression level of MMP- 2 in the colon adenoma was higher than that in the normal colonic tissues （P〈0.05）. The expression level of MMP- 2 and MMP- 7 in the colon cancer was higher than that in the colon adenoma and normal colonic tissues（P〈0.05）. In colon cancer tissues, the expression level of MMP- 2 and MMP- 7 in stage Ⅲ and Ⅳ was higher than that in stage Ⅰ and Ⅱ（P〈0.05）. The expression level of MMP- 2 and MMP- 7 in stage Ⅳ was higher than that in stage Ⅲ （P〈0.05）. Conclusions： The expression level of MMP- 2 and MMP- 7 is related with the formation,invasion and translation of colon neoplasm. The mechanism of higher expression need more exploration.
