摘要 目的观察β-半乳糖苷酶(β-gal)在ROSAβgeo26小鼠衍生的神经干细胞(NSCs)中的表达变化。方法体外培养NSCs,并应用X-Gal组织化学和免疫细胞化学染色技术研究NSCs中β-gal的表达变化。结果原代克隆球和亚克隆球的NSCs均稳定表达β-gal;在NSCs分化过程中,NeuN阳性的神经元、GalC阳性的少突胶质细胞和GFAP阳性的星形胶质细胞均表达β-gal,但表达水平有所下降。结论ROSAβgeo26小鼠衍生的NSCs能稳定的表达β-gal,可用作NSCs研究的细胞来源。但在干细胞分化的相关研究中,值得注意存在β-gal不同程度的表达下调现象,并应进一步关注出现该现象的潜在机制。 Objective To observe the expression of β-galactosidase (β-gal) in the neural stem cells (NSCs) of ROSAβgeo26 mouse. Methods NSCs were cultured in vitro. Then X-Gal histochemistry and immunocytochemistry were performed to study the expression of β-gal in the NSCs. Results β-gal was stably expressed in the primary clones and subclones of NSCs. During the differentiation of NSCs, β-gal activity was colocalized to NeuN positive neurons, GalC positive oligodendrocytes and GFAP positive astrocytes, but the level of β-gal expression was down-regulated. Conclusion β-gal is stably expressed in the NSCs of ROSAβgeo26 mouse. This strain of mice may serve as a promising resource for study of NSCs. But in the research related to NSCs differentiation, β-gal down-regulated expression of raring degrees and its underlying mechanism should be noted.
