脐血单个核细胞IL-12和IFN-γ表达的观察

摘要 本文探讨新生儿脐血单个核细胞(CBMC)白细胞介素12(IL-12)和γ-干扰素(IFN-γ)基因表达水平;观察重组IL-12(rIL-12)对CBMCIFN-γ基因表达的影响。采用逆转录-聚合酶链式反应(RT-PCR)测定8例新生儿CBMCIL-12p_(40)mRNA和IFN-ymRNA表达,用ELISA法测新生儿CBMC细胞培养上清液中IFN-γ浓度。结果显示:新生儿CBMC在体外受刺激后表达的IL-12p_(40)mRNA、IFN-γmRNA均较成人PBMC低下;新生儿CBMC培养上清液中IFN-γ水平明显低于成人,rIL-12能增强PMA和抗CD_3单克隆抗体(McAb)刺激的新生儿CBMC产生IFN-γ,使其达到成人水平。提示:缺乏IL-12调节作用可能是新生儿CBMCIFN-γ基因表达水平不足的重要原因之一。 The objective of this study was to explore the levels of interleukin 12(IL-12) and IFN-γ gene expression in neonatal cord blood mononuclear cells (CBMC). The expression of IL-12 p40 mRNA and IFN-γ mRNA in neonatal CBMC of 8 cases were determined with reverse transcription-polymerase chain reaction (RT-PCR),and the concentration of IFN-γ in supernatant of cultured neonatal CBMC was determined with enzyme linked immunosorbent assay (ELISA). The results showed that the expression of IL-12p40 mRNA and IFN-γ mRNA in CBMC were lower than those of adults PBMC after being stimulated in vitro,the level of IFN-γ in supernatant of cultured neonatal CBMC was significantly lower than that of adults, rIL-12 could enhance IFN-γ producing in CBMC stimulated by PMA and anti-CD3 monoclonal antibody up to the level of adults. The results of this study suggest that lack of regulation by IL-12 may be an important cause of insufficient IFN-γ gene expression in neonatal CBMC.
