摘要
目的观察δ氨基酮戊酸(δ-aminolaevulinic acid,ALA)诱导人结肠癌细胞SW480内产生内源性卟啉IX(protopor-phyrin IX,PPIX)规律以及与光动力学疗法(photodynamic therapy,PDT)杀伤作用的关系。方法在SW480结肠癌细胞培养的基础上,采用激光共聚焦显微镜分析ALA诱导SW480结肠癌细胞内产生PPIX的变化规律;用MTT法检测细胞存活率,研究杀伤作用规律。结果加入ALA后SW480细胞产生PPIX的量随时间延长,PPIX量逐渐增加,加药后8h达最高峰,24h后恢复到正常细胞水平。PPIX荧光主要产生在细胞浆内,细胞核内较少。PDT24h后细胞存活率开始明显降低,PDT杀伤作用与癌细胞内PPIX量呈负相关。结论ALA诱导SW480细胞产生PPIX的量在加药后8h达最高峰,24h后恢复到正常细胞水平,PPIX荧光主要分布在细胞浆内。PDT杀伤作用与癌细胞内光敏剂明显相关。
Objective To investigate protoporphyrin IX(PPIX) level induced by δ-aminolaevulinic acid(ALA) in SW480 colonic adenocarcinoma cell and the relationship between PPIX level and effects of photndynamic therapy(PDT) on SW480 colonic adenocarcinoma cell. Methods On the basis of cell culture,laser confocal fluorescence microscopy and MTT assay were applied to investigate the intracellular fluorescence intensity induced by ALA and surviving cell fraction in photodynamic therapy of SW480 colonic adenocarcinoma cell. The results were analyzed with SPSS 10.0 statistical software. Results Fluorescent intensities of PPIX became much stronger with prolonging time of ALA exposure. Maximum PPIX accumulation in the SW480 cell was seen at 8h after exposure of ALA. Fluorescent intensities of PPIX would recovery normal leval at 24h after exposure of ALA. Much flourescence of PPIX was distributed in endochylema,less in nucleus. The cell surviving fraction of SW480 was significantly decreased after PDT for 24h,compared with that of control group(P〈0.01). The killing effect of PDT on SW480 colonic adenocarcinoma cell was negatively correlated with intracellular PPIX level. Conclusion Maximum PPIX accumulation in the SW480 cell is seen at 8h after exposure of ALA. Intracellular PPIX will be metabolized completely in 24h after administration of ALA. The killing effect of PDT to SW480 cells is related to intracellular PPIX level.
