摘要
采用随机扩增多态性DNA(RAPD)分析和核糖体(rDNA)内转录间隔区(ITS)序列分析方法对14种曲霉进行分类鉴定与系统发育研究。RAPD分析构建的聚类树状图,显示出不同的曲霉种间只在较低的相似性系数聚为一类,这与形态特征分种基本吻合,可以清楚地将不同的种区分开来。ITS序列分析结果表明,形态上相似的6种曲霉之间的绝对遗传距离大于11个碱基,属于种间的差别范围。对相同的供试菌株两技术所反映亲缘关系的趋势相同,也表明了这两种技术用于曲霉亲缘关系分析的种类鉴定具有合理性和可行性。
Random amplified polymorphic DNA (RAPD) markers and the DNA sequences of ribosomal DNA internal transcribed spacer (ITS) were used to analyze the relationships of 14 Aspergillus species, in the cluster analysis of the RAPD bands, the result showed that isolates of different species were clustered in low value of similarity coefficient by Between-groups linkage and similarity matrix.This result was almost consistent with the morpholog- ical studies, and also indicated the rationality and feasibility of the application of RAPD analysis in the identification of Aspergillus species. This result of ITS sequence analysis showed that the absolute distance among of them was over 11 base pairs, which was the normal range of different species.This result indicated the rationality and feasibility of the application of RAPD analysis and rDNA ITS sequences in the identification of Aspergillus species.
