α-乳香酸对HL-60细胞株VEGF分泌及其Flt-1受体表达的影响 被引量:7

Effect of a-boswellic Acid on VEGF and Its Receptor Expression in Acute Promyelocytic Leukeomia Cell Lines HL-60 Cells

摘要 [目的]探索不同浓度α-乳香酸作用于白血病细胞株HL-60细胞末同时间后,细胞血管内皮生长因子(VEGF)mRNA表达及VEGF蛋白分泌及受体Flt-1的变化。[方法]MTT法检测乳香酸对细胞增殖的抑制作用,选择适当的药物浓度及作用时间;RT-PCR法检测α-乳香酸处理前后HL-60细胞中VEGFmRNA表达水平;流式细胞术检测α-乳香酸处理前后细胞中VEGF和受体Flt-1蛋白表达水平。[结果]α乳香酸在15.0mg/L时在体外能下调HL-60细胞VEGFmRNA的表达和VEGF蛋白的分泌并呈时间与剂量依赖性(P〈0.05)。[结论]α-乳香酸在体外可以抑制急性早幼粒细胞白血病血管新生,其作用可能与下调VEGF及Fit-1有关,具有抗白血病效应。 [Objective] To assess the effects of α-boswellic acid on VEGF and Flt-1 expression in acute promyeiocytie leukemia cell Lines HL-60 cells. [Methods] HL-60 cells were treated with α-boswellic acid, then cell proliferation was examined by the method of MTT. The level of VEGF-mRNA expression was distin- guished by semi-quantitative RT-PCR technique. Flow cytometry was applied to examine the protein expression of VEGF and Flt-1. [Results] Within the concentration of (10.0-15.0)mg/L, α-boswellic acid strongly inhibited proliferation of HL-60 cells. When the drug was 15.0mg/L, α-boswellic acid inhibited the mRNA expression of VEGF and the protein expression of VEGF and Flt-1. [Conclusion] Within a certain drug concentration, α-boswellic acid can inhibit the proliferation of HL-60 cells. The role of inhibiting angiogenesis maybe relates with down regulating of the expression of VEGF and Fh-1.
