摘要
目的观察血肿周边脑组织中HIF-1α与VEGF蛋白的表达情况并探讨二者表达的相关性。方法SD大鼠35只,随机分为对照组、假手术组、脑出血组,脑出血组又分为6h、24h、72h、7d和21d5个亚组,每组5只。采用自体血脑内注射法建立脑出血动物模型,应用免疫组化的方法检测脑出血后不同时间血肿周边脑组织中HIF-1α与VEGF蛋白的表达情况。结果脑出血后,HIF-1α和VEGF蛋白的表达均随时间呈进行性增加,并于出血后24~72h达到高峰且维持到第7天。与对照组和假手术组比较,差异均有统计学意义(P<0.01)。脑出血后,HIF-1α和VEGF蛋白的表达呈正相关。结论HIF-1α与VEGF蛋白在血肿周边脑组织中均有表达,并具有一致性,可能与脑出血后微循环的重建有关。
Objective To investigate the expression of HIF-la and VEGF after intracerebral hemorrhage (ICH) and to find out their eharacters and correlation. Methods The model of ICH was established by injecting 50μl of autologous blood into the right caudate putamen. Thirty-five healthy SD rats were randomly divided into normal group(n= 5), sham-operated group (n=5) and ICH model group (n=25). Rats were sacrificed at 6h,24h,72h,Td and 21d after operation, respectively. The expressions of HIF-1α and VEGF were investigated by using SP immunohistochemistry. Results The positive cells of HIF-1α and VEGF increased in model group while no positive cells in normal group and sham-operated group(P〈0.01). The peak value appeared at 24 hours to 72 hours in model group. There was positive correlation between HIF-1α protein and VEGF protein by Pearson's correlation coefficient(r=0. 735,P〈0. 01). Conclusion HIF-1α protein and VEGF protein were over expressed in brain tissues and there was significant correlation after ICH. It can probably help angiogenesis in the condition of hypoxia.
