摘要
[目的]寻找3种观赏凤梨花蕾组织培养的适宜培养基,建立组培快繁体系。[方法]以3种观赏凤梨的花蕾作为外植体进行组织培养,找出合适的诱导培养基、增殖培养基和生根培养基。[结果]确定3种观赏凤梨花蕾的最佳诱导培养基为MS+6-BA3.0 mg/L+ZT1.5 mg/L+NAA0.5 mg/L,增殖的适宜培养基为MS+6-BA1.0 mg/L+NAA0.1 mg/L,生根的适宜培养基是1/2MS+IBA0.3 mg/L+NAA0.2 mg/L+活性炭0.5 g/L。[结论]以观赏凤梨花蕾为外植体的组培过程无褐化现象产生,且伤害母株程度小,污染率低。该研究结果为生产企业的大规模种苗生产提供了强有力的技术支持。
[Objective] The aim was to seek the optimum medium for the tissue culture of three kinds of ornamental pineapple flower buds and establish the tissue culture and rapid propagation system. [Method] Three kinds of omamental pineapple flower buds were used as explants to make the tissue culture and find out the suitable induction medium, proliferation medium and rooting medium. [Result] The optimum induction medium for three kinds of ornamental pineapple flower buds was confirmed as MS + 6-BA 3.0 mg/L + ZT 1.5 mg/L + NAA 0.5 mg/L. The optimum proliferation medium was MS + 6-BA 1.0 mg/L + NAA0.1 mg/L and the optimum rooting medium was 1/2 MS + IBA0.3 mg/L + NAA0.2 mg/L +active carbon 0.5 g/L. [Conclusion] There were no browning phenomenon, little harm on the mother plant and low pollution rate in the tissue culture process with ornamental pineapple flower buds as explants. The research results provided a powerful technical support for large-scale seedlings production of production enterprises.
