转化生长因子β亚型及其受体在人类系膜增生性肾小球肾炎各期病变表达的研究 被引量:16

摘要 目的研究人系膜增生性肾小球肾炎(MsPGN)病变进展过程中转化生长因子β(TGFβ)各亚型及其受体的变化。方法应用免疫酶染色技术,检测人轻度MsPGN、中度MsPGN及重度MsPGN时,肾组织中TGFβ各亚型及其受体及细胞外基质成分(Ⅳ型胶原、纤粘连蛋白、层粘连蛋白及Ⅲ型胶原)的表达情况。结果在轻度MsPGN时,肾小球中无TGFβ1、β2、β3及其受体(TβRI)的表达。中、重度MsPGN时,上述TGFβ各亚型及其受体在肾小球中表达并增加。同时肾小球中细胞外基质各成分的含量也显著增加。 Objective To observe the role of transforming growth factor (TGF)β isoforms and their receptor in the development of human mesengial proliferative glomerulonephritis (MsPGN). Methods Immuno histochemical staining (LAB) method was used to detect the expression of TGFβ isoforms (TGFβ 1, β 2, β 3), type Ⅰ receptor (TβRI), and deposition of collagen Ⅳ, collagen Ⅲ, fibronectin, and laminin. Results In mild MsPGN group, glomeruli showed no immunostaining for all three TGFβ isoforms and TβRI, but TGFβ 1 expressed a little in interstitium and TGFβ 2, β 3 and TβRI also a little in renal tubules. In moderate and severe MsPGN groups, however, TGFβ 1, β 2, β 3 and TβRI emerged and increased in glomeruli. Meanwhile glomerular deposition of four matrix components (collagen Ⅳ, collagen Ⅲ, fibronectin and laminin) was also significantly increased with the progression of MsPGN, revealing a close relationship between the increased expression of all the three TGFβ isoforms and severity of the disease. The expression of TGFβ 1 in interstitium and TGFβ 2, β 3 and TβRI in renal tubules was also increased. Conclusion Increased expression of all the three TGFβ isoforms and TβRI is associated with pathological accumulation of extracellular matrix in MsPGN.
