摘要
应用细胞因子生物学检测法对IL-12、IL-7、IL-2协同诱导的PBMC培养上清中的TNF和IL-2进行了检测,同时观察了抗TNF-α、抗TNF-β和抗IL-2抗体对刺激细胞的细胞毒作用的影响。结果表明,IL-12、IL-7、IL-2三种因子协同刺激PBMC可产生较高水平的TNF,而抗TNF-α单抗能显著抑制刺激细胞的杀瘤活性,提示内源性TNF-α在介导该类细胞的细胞毒效应中起重要作用。
The bioassay were used to estimate the level of TNF and IL2 in the supernatant of PBMC induced by IL12,IL7 and IL2.The effect of antiTNFα,antiTNFβ and antiIL2 antibody on cytotoxicity of PBMC were detected.The results showed that synergistic stimulation of IL12,IL7 and IL2 could produce high concentration of TNF in culture supernatant,and antiTNFα could obviously inhibit the cytotoxic activity of treated PBMC,therefore,the endogenous TNFα was considered playing important role in the cytotoxic activity of treated PBMC.
