摘要
以卡瑞(Carrez)试剂为样品净化剂,采用聚焦微波萃取系统(FMASE)提取水果中的多酚类物质,建立了水果中8种常见多酚化合物的反相高效液相色谱(RP-HPLC)分析方法,比较和探讨了样品的微波提取法与常规榨汁法。结果表明,微波提取法中各组分均能得到基线分离,标准曲线相关系数均达0.999以上,加标回收率为90%-110%,相对标准偏差小于3.6%。对梨、苹果及香蕉的测定结果表明,果皮比果肉含有的多酚种类多、含量相对较高;梨皮中芦丁和绿原酸含量相对较高;香蕉皮中儿茶素含量最高。应用微波提取制样法较常规榨汁法检测到更多种的多酚类物质。
A novel method was developed for extraction and determination of poly-phenols in fruit. The poly-phenols in the periearp and pulp of fruit were extracted with focused microwave-assisted solvent extraction (FMASE) by using 70% methanol under the microwave power of 200 for 15 min. The extract treated with Carrez reagent was analyzed by a reversed-phase high performance liquid chromatography (RP-HPLC) using 0.01mol.L^-1 methanol- phosphoric acid solution as the mobile phase and 280 nm as the UV detection wavelength. The calibration curves for the eight poly-phenols showed a good linearity with the correlation coefficients of more than 0. 999. The spiked recoveries were range of 90% - 110% and the relative standard deviations were within 3.6%. The quantitative results showed that the kinds and content of poly-phenols in pericarp were more than those in pulp. The comparison of two extraction methods also indicated that the extraction efficiency of microwave extraction was higher than that of expression extraction, and the more kinds of poly-phenols could be detected by using microwave extraction method.
