摘要
通过高效液相色谱法对传统中药黄连中的主要成分小檗碱的含量进行测定.使用kromasil C18色谱柱(250 mm×4.6 mm,5μm)以乙腈和水作为流动相进行梯度洗脱流速1 mL/min,检测波长为345 nm.盐酸小檗碱浓度在5μg/mL~50μg/mL范围内线性关系良好,平均加样回收率为99.56%,RSD为2.75%.本方法简便、准确、快速、重复性好,可用于黄连中小檗碱的含量测定.
To establish a HPLC method for the determination of Berberine in Coptis chinensis F ranch.LC separation was achieved using a kromasil C18(250 mm×4.6 mm,5 μm) maintained at 30 ℃.The mobile phase consisted of A(acetonitrile) and B(0.2% acetic acid and 10 mmol·L-1 ammonium acetate) with linear gradient elution.The detect wavelength was 345 nm.The flow rate was 1 mL/min during the whole gradient cycle.The linear concentration range was from 5 μg/mL~50 μg/mL.The average recovery was 99.56%,RSD=2.75%.The method is simple,accurate,rapid and reproducible.It can be used to detect the content of berberine.
