摘要
建立了高效液相色谱-蒸发光散射检测法(HPLC-ELSD)同时测定盐酸大观霉素中糖的新方法.乙腈-水为流动相,流速1.0 mL/min,柱温25℃,蒸发光散射检测器漂移管温度80℃,氮气作载气,流速2.00 L/min.糖的线性范围:果糖、葡萄糖、蔗糖、麦芽糖0.5-4.5μg,相关系数为0.999 1-0.999 8,四种糖的加标回收率范围为98.8%-109.8%,检出限(S/N=3)低于3μg/mL.
A new approach for simultaneously determination of carbohydrates in spectionmycin hydrochtoride by High Performance Liquid Chromatography with Evaporative Light Scattering Detector (HPLC-ELSD) was developed. The carbohydrates were separated using an acetonitrile-water gradient system at a flow-rate of 1.0 mL/min. The optimum setting of the ELSD parameters:drift tube temperature was 80% and nitrogen carrier gas flow-rate was 2.00 mL/min. The ab of the carbohydrates(fructose, glucose, sucrose, maltose)showed distinct linearity relation ship with the concentration of the carbohydrates in the range of 0.5 - 4.5μg, the correlation coefficient was 0. 9991 - 0.9998. Recovery of 98.8 % - 109.8% was obtained, and the detection limit(S/N = 3) was found to be under 3μg/ml.
