摘要
目的:研究Sertoli细胞对体外培养大鼠皮质神经前体细胞生长发育的促进作用,及向多巴胺能神经元分化的诱导作用。方法:将大鼠Sertoli细胞与14d大鼠皮质神经前体细胞体外共培养,免疫细胞化学检测神经干细胞标记物巢蛋白,神经元、星形胶质细胞和小胶质细胞标记物βⅢ-微管蛋白(βⅢ-tubulin)、胶质纤维酸性蛋白(GFAP)和半乳糖脑苷脂(GalC),及多巴胺能神经元标记物酪氨酸羟化酶(TH)的表达情况。结果:随着共培养时间的延长,神经前体细胞中GFAP阳性细胞数量逐渐减少,而β-Ⅲtubulin阳性细胞数增多,并出现TH阳性神经元。结论:Sertoli细胞抑制体外培养的大鼠皮质神经前体细胞的生长,但具有显著促神经元分化作用,并能够诱导与其共培养的大鼠皮质神经前体细胞分化为多巴胺能神经元。
To investigate the effect of Sertoli cells from the testis on proliferation and differentiation of cultured neural precursors (NPCs) derived from the cerebral cortex in rats. Methods: Sertoli ceils of Wistar rats were co-cultured with NPs. The detections of β-Ⅲ tubulin,GFAP, GalC and tyrosine hydroxylase (TH) were made by immunocytochemistry in vitro. Results: The numbers of NPCs with nestin-positive was decreased with increasing co-culture time, so were the GFAP and GalC. β-Ⅲ tubulin-positive cells increased, and TH-positive cells appeared in the co-culture. Conclusion: Sertoli cells exert the negative effect on the proliferation of NPCs, but have distinctly positive effect on NPCs for its differentiation into neurons in vitro. The neural precursors from the cortex of rats can be directly differentiated into the dopaminergic neurons when co cultured with the Sertoli ceils.
