摘要 目的:观察二氧化钛(TiO2)纳米颗粒随时间变化对小鼠单核细胞超微结构的影响。方法:将50μg/ml纳米TiO2颗粒溶液与小鼠单核细胞(RAW264.7)共培养,在37℃,5%CO2的细胞培养箱中分别孵育1、2、4、8、16、24h,收集细胞,透射电镜下观察小鼠单核细胞超微结构的改变。结果:纳米TiO2的平均粒径为30nm,当TiO2纳米颗粒与单核细胞作用4h以下时,细胞中有少量纳米TiO2颗粒,线粒体和内质网出现不同程度的肿胀或扩张,细胞核无明显变化。随着作用时间的增加,细胞核也出现不同程度的变化。讨论:纳米TiO2颗粒以胞吞的方式进入细胞,对各细胞器产生影响,其中主要作用于细胞核。 Objective: To observe the effect of nano sized TiO2 in mouse monocyte cells with increasing incubation time. Methods: Mouse monocyte cells were incubated for 1, 2, 4, 8, 16 and 24 h with nano-sized TiO2 at 50 bμg/ml in cell incubater with 37℃C and 5% CO2. After preparing the sample, we observe the change of ultrastructure of mouse monocyte-macrophage cells. Results: Treated with 50/μg/ml nano-sized TiO2 (30 nm in mean diameter), a small amount of nanc-particles in cells, and some changes of cell ultrastructure were observed. Different degrees of change appeared in the nucleus with increasing incubation time. Conclusion: Nano-TiO2 particles entered monoeyte cells by endocytosis, and affected various organelles, especially nucleus.
