白细胞介素-1β体外诱导小鼠中脑神经干细胞向酪氨酸羟化酶阳性神经元分化 被引量:1

摘要 目的:探讨白细胞介素-1β(IL-1β)体外诱导中脑神经干细胞(M-NSCs)向酪氨酸羟化酶阳性神经元分化情况,为帕金森病的转基因移植治疗在体外获得足够的多巴胺能神经元提供实验依据。方法:在有血清条件下体外培养鼠胚M-NSCs,予以IL-1β作诱导分化。酪氨酸羟化酶(TH)免疫细胞化学鉴定,流式细胞术检测TH阳性神经元的百分率。结果:实验组促进M-NSCs分化为TH阳性神经元的比例为9.54%,而对照组仅3.46%,差异有统计学意义。结论:IL-1β可明显促进中脑神经干细胞分化成多巴胺能神经元。 Objective: To explore the feasibility of differentiation of mice mesencephalic neural stem cells (M-NSCs) into do paminergic positive cells induced by IL-1β, and provide the experimental basis for cell therapy of Parkinson disease. Methods: After the M-NSCs were isolated and harvasted, IL-1 beta was added into the culture media supplementing with fetal boving serum. Immunocytocbemical staining was applied to identify the TH positive cells and was used to define the induction efficiency. Results: After induction, about 9.54% MNSCs were shown positive reaction to TH, with stastical difference compared to control ones, which occupying 3.46% positive. Conclusion: IL-1β can promote significantly the differentiation of M-NSCs into DAergic neurons.
