SEZ6基因在人肿瘤细胞的表达

目的通过检测SEZ6基因在15种不同组织来源的人肿瘤细胞中的表达情况,以探讨SEZ6基因与肿瘤的发生、发展之间的关系,为进一步研究SEZ6基因的生物学功能提供理论依据。方法体外培养15种不同组织来源的人肿瘤细胞,抽提总RNA,运用RT-PCR检测SEZ6基因的表达情况。结果(1)SEZ6在肿瘤细胞K562、A2780、SHP-77、H1299、SGC 996、A549、H2122、SPCA、MHCC-97H、SW480、MDA-MB-435中表达,其中,在A2780、H1299、A549、MHCC-97H、SW480及MDA-MB-435细胞系中高表达,与阳性对照组无差异。另外,在6种肺癌细胞系中,5种(SHP-77、H1299、A549、H2122、SPCA)有SEZ6基因表达;(2)在高转移性肿瘤细胞MHCC-97H、SW480、MDA-MB-435、H1299和SHP-77中SEZ6高表达,与低转移性肿瘤细胞有显著性差异,P<0.05。结论SEZ6基因在部分肿瘤细胞中表达,并在高转移性肿瘤细胞中高表达,提示SEZ6基因可能参与了肿瘤的发展过程,并与肿瘤的转移有关。

Objective To study the function of seizure related gene 6 （SEZ6） and the relationship between SEZ6 gene and the occurrence, development of tumor by detecting the expression of SEZ6 gene in 15 kinds of human cancer cell lines. Methods Total RNA was extracted from fifteen kinds of human cancer cell lines which had been cultured in vitro. The expression of SEZ-6 was detected by RT-PCR in these cell lines. Results （1） The expression of SEZ6 was detected in cell lines of K562, A2780, SHP-77, H1299, SGC 996, A549, H2122, SPCA, MHCC-97H, SW480 and MDA-MB-435. SEZ6 was over-expressed in cell lines of A2780, H1299, A549, MHCC-97H, SW480 and MDA-MB-435, which had no difference compared with positive group. SEZ6 gene was expressed in 5 lung cancer lines out of 6 being researched. （2） Compared with lowly metastatic cell lines, there were obviously high expression in highly metastatic ones such as MHCC-97H, SW480, MDA-MB-435, H1299 and SHP-77 （P 〈 0.05）. Conclusion SEZ6 gene expresses in partial human cancer cell lines, especially in highly metastatic cancer cells. It seems that this gene plays a role during the occurrence and development of tumor and it has some relationship with metastasis of tumor.
