摘要
目的研究羟丙基-β-环糊精(HP-β-CD)及其包合物的超滤适用性。方法以温度、压力、浓度、超滤膜截留分子质量4因素进行正交设计,成分含量为检测指标,测定超滤前后药液成分的浓度,计算透过率来观察HP-β-CD及其包合荆芥挥发油和丹皮酚溶液超滤前后的含量变化。结果HP-β-CD不被膜吸附,透过率高;荆芥挥发油及丹皮酚的HP-β-CD包合物均能被膜吸附,透过率高;超滤膜相对截留分子质量因素对荆芥挥发油包合物的透过率有显著影响(P<0.05);浓度因素对丹皮酚包合物的吸附率有非常显著影响(P<0.01)。结论HP-β-CD在超滤过程中溶液浓度不发生变化,荆芥挥发油及丹皮酚的HP-β-CD包合物在超滤过程中因被膜吸附而有一定损失。
AIM To investigate the applicability of ultrafihration of hydroxypropyl-β-cyclodextrin (HP-β- CD) and its inclusion compound. METHODS Component content was selected as detecting indicator. Temperature, pressure, concentration, and cut off molecular weight (MWCO) of ultrafihration membrane were selected as factors for orthogonal design method, the concentration was determinated in drug solution before and after using the ultrafiltration, and the transmittance was calculated to detect the content variation of HP-β-CD and its inclusion compound of schizonepeta volatile oil and paeonol solution. RESULTS HP-β-CD was not absorpted by the membrane and had high transmittance. The inclusion compound of HP-β-CD of schizonepeta volatile oil and paeonol were absorpted by the membrane but had high transmittance. MWCO had a significant effect on transmittance rate of schizonepeta volatile oil (P 〈 0.05). Concentration had a very significant effect on absorbance rate of paeonol (P 〈 0.01). CONCLUSION Uhrafiltration had little effect on the concentration of HP-β-CD but its inclusion compound of schizonepeta volatile oil and paeonol solution had some lost.
