摘要
目的:调查分析广州地区泌尿生殖道感染沙眼衣原体隐蔽性质粒的分子流行病学。方法:设计合成隐蔽性质粒特定序列区域的特异性引物,半巢氏PCR扩增沙眼衣原体隐蔽性质粒DNA片段,同时结合omp1基因PCR扩增,比较分析质粒型。结果:143例沙眼衣原体ELISA检测阳性的标本,隐蔽性质粒内外引物扩增全部阳性,而omp1基因PCR扩增检出140例阳性。质粒分型为质粒完整型沙眼衣原体143株菌,未见质粒缺失型和无质粒型菌株。结论:广州地区尚未发现质粒变异型沙眼衣原体,有效地检测沙眼衣原体质粒分子流行病学状况对于实验室诊断和制定控制策略有重要的意义。
Objective: To investigate the molecular epidemiology of cryptic plasmid of Chlamydia trachomatis isolated from urogenital samples from STD patients in Guangzhou. Methods:Specific primers were designed and synthesized according to DNA sequences of the target region of cryptic plasmid and ompl gene. The special sequence of the plasmid DNA were amplified by semi -nested PCR. Compared with results of the ompl gene amplification, types of cryptic plas- mids were analyzed. Results:Out of 143 ELISA - positive samples, 143 samples were positive cryptic plasmid by inner and outer primers, and 140 samples were positive for ompl gene. Typing of Chlamydia trachomatis in 143 samples showed all of them had intact crypt plasmids. Conclusion: No chlamydia trachomatis variant were found in Guangzhou at the time of this investigation. Exploration of molecular epidemiology of cryptic plasmid of Chlamydiatra chomatisin may prove critical in order to improve strategies for diagnosis and disease control.
