摘要 目的研究金属蛋白酶解离素28(ADAM28)反义核酸(AS-ODN)对人牙乳头细胞(HDPC)增殖、分化、凋亡等生物学特性的影响并分析可能的作用机制。方法采用细胞培养、基因转染、四唑盐(MTT)比色法、酶动力学法和流式细胞术(FCM)检测ADAM28反义核酸转染HDPC后对细胞生物学特性的影响。结果ADAM28AS-ODN组HDPC的增殖活性、增殖指数、碱性磷酸酶(ALP)分泌活性显著降低,凋亡细胞百分比明显上升。结论ADAM28反义核酸可显著抑制HDPC的增殖和ALP的活性,显著诱导HDPC的凋亡并影响细胞周期的变化。 Objective To study the effects of a disintegrin and metalloproteinase 28 (ADAM28) antisense oligodeoxynucleotide(AS-ODN) on proliferation,differentiation and apoptosis of human dental papilla cells(HDPC) and analyze its possible mechanism. Methods Cell culture, gene transfection, MTT chromatometry, enzyme dynamics, and flow cytometry (FCM) techniques were used to examine the effects of ADAM28 AS-ODN on the biological characteristics of HDPC after HDPC was transfected by ADAM28. Results In ADAM28 AS-ODN group, proliferation activity, proliferation index and alkaline phosphatase (ALP) secretion activity of HDPC decreased significantly, and percentage of cell apoptosis increased obviously. Conclusion ADAM28 AS-ODN could inhibit proliferation and ALP activity of HDPC, induce apoptosis of HDPC and influence the changes of cell cycle.
