摘要
目的:研究血红素氧合酶1(HO-1)、血红素氧合酶2(HO-2)在雌性去势大鼠阴道平滑肌中的表达,探讨雌激素对阴道平滑肌HO-1、HO-2表达的影响。方法:雌性8周龄SD大鼠随机分为A(去卵巢2周组)、B(去卵巢4周组)、C(假手术2周组)、D(假手术4周组)4组,每组6只,放免法测定各组雌性大鼠血清雌二醇水平,运用免疫组化和RT-PCR对HO-1、HO-2在阴道平滑肌进行定位和半定量分析。结果:术后A与C、B与D组血清雌二醇比较有显著差异(P<0.05);HO-1、HO-2免疫组化定位在阴道平滑肌细胞胞质内,A组表达显著低于C组;阴道平滑肌组织中HO-1、HO-2mRNA表达在A、B组均显著低于C、D组,阴道中HO-1mRNA、HO-2mRNA下降与血清雌二醇下降相关。结论:HO-1、HO-2在雌性大鼠阴道平滑肌中有表达,低水平的雌激素与HO-1、HO-2表达降低有关,雌激素可能经HO/CO途径在阴道平滑肌中发挥生理作用。
Objective : To analyze the changes of heme oxygenase-1 ( HO-1 ) and heme oxygenase-2 ( HO-2 ) in the vaginal smooth muscle of ovarieetomized female rats and to investigate the effects of estrogen on the expressions of HO-1 and HO-2 in the vaginal smooth muscle. Methods: Twenty-four 8-week old female SD rats were equally randomized into Group A (2-week castrated), B (4- week castrated), C (2-week sham operation) and D (4-week sham operation). The levels of plasma estradiol were measured by radioimmunoassay for all the rats and the expressions of HO-1 and HO-2 in the vaginal muscle were detected and analyzed by immunohistochemistry and RT-PCR. Results : The postoperative level of plasma estradiol was significantly lower in Groups A and B than in C and D (P 〈 0.05 ). And the expressions of HO-1 and HO-2 in the vaginal smooth muscle were significantly decreased in the first two groups as compared with the latter two. Conclusion: HO-1 and HO-2 are expressed in the vaginal smooth muscle of the female rats, and their decreased expressions are correlated with the lowered level of plasma estradiol. Estrogen may play a physiological role in the vaginal smooth muscle via HO/CO.
