摘要
目的检测超排卵小鼠着床期子宫内膜白血病抑制因子(leukemia inhibitory factor,LIF)的表达水平,探讨补肾安胎方对超排卵小鼠着床期子宫内膜容受性的影响。方法将120只小鼠随机分为正常组、模型组、中药组。模型组和中药组注射PMSG和HCG造成小鼠超排模型后合笼,正常组注射等体积生理盐水后合笼。中药组给予补肾安胎方,正常组和模型组灌服等量蒸馏水。于妊娠第4天(Pregnancy day4,Pd4)、第五天(Pd5)留取妊娠子宫;采用免疫组化技术检测LIF蛋白表达,采用RT-PCR技术检测LIF mRNA表达。结果免疫组化SP法检测结果显示3组小鼠LIF蛋白表达空间分布无明显差异。RT-PCR技术检测结果显示正常组Pd4LIF表达呈强阳性,Pd5表达下降,与模型组同期比较差异有统计学意义(P<0.05),中药治疗后与模型组同期比较差异有统计学意义(P<0.05)。结论补肾安胎方能协调由于控制性超排卵所致的子宫内膜LIF表达水平,从而提高小鼠着床期子宫内膜容受性。
Objective To detect the expression of leukemia inhibitory factor (LIF) mRNA and protein, and to study the impact of Bushen Antai Recipe(BAR) on endometrial receptivity in Controlled Ovarian Hyperstimulation (COH) mice during the stage of implantation window. Methods 120 kunming mice were randomized into three groups:normal control group, model group and Traditional Chinese Medicine group (TCM group). The mice in the model and TCM groups were kept in a cage for mating after being injected with PMSG and HCG for inducing controlled ovarian hyperstimulation. The mice in the normal control group were kept in a cage for mating after being injected with saline. The mice in the TCM group were administered with BAR and the mice in the normal and model groups were given distilled water. Uterus were collected on the pregnancy day 4 (Pd4) ,5 mice post coition. The expression of LIF protein and mRNA were detected by immunohistochemistry and RT-PCR respectively. Results Immunohistochemistry observation showed that there was no remarkably different in spatial distribution between the three group. RT- PCR observation showed that On Pd4, the expression of LIF mRNA in the normal control group was the highest, and then gradually depressed on Pd5,which has remarkable difference from that in the model group(P 〈0.05 ). The expression of LIF mRNA in the TCM group is also markedly different from the model group ( P 〈 0.05 ). Conclusion It was suggested that BAR could improve endometrial receptivity of mice with COH by promoting the expression of LIF.
