摘要
目的观察益气活血方及其拆方对出血性血小板病患者凝血酶受体血小板膜糖蛋白(GP)Ⅰb/Ⅸ/Ⅴ复合物及其组分GPⅠbα的表达的影响来探讨其配伍规律。方法68例出血性血小板病患者及32例健康人静脉血在静息状态及体外加益气活血方及其拆方中药孵育后,采用流式细胞术检测血小板GPⅠb/Ⅸ/Ⅴ及GPⅠbα的表达。结果静息状态时,患者GPⅠb/Ⅸ/Ⅴ及GPⅠbα的表达明显低于健康人;益气活血方及其部分拆方组能使患者凝血酶受体GPⅠb/Ⅸ/Ⅴ及GPⅠbα恢复至正常水平;而单味中药组作用不明显;方中的主药黄芪与当归配伍为用,可促进血小板膜糖蛋白的表达。结论益气活血全方及其部分拆方组在受体蛋白水平上调节GPⅠb/Ⅸ/Ⅴ及GPⅠbα的表达,是其治疗出血性血小板病的疗效机制之一。
Objective To investigate and explain the composing mechanism by observing effect of Yiqi Huoxue decoction (YQHXD) and its decomposed formulas on expression of platelet membrane glycoprotein (GP) Ⅰb/Ⅸ/Ⅴ complex and GPⅠbα in patients with hemorrhagic thrombopathy (HTP). Methods Venous whole blood was obtained from 68 patients with HTP and 32 healthy human subjects, GPⅠb/Ⅸ/Ⅴ and GPⅠbα levels on the membrane of stationary platelet or platelet by in vitro incubation with YQHXD and its decomposed formulas were measured with using cytometric method. Resuits The mean fluorescence intensity of GPⅠb/Ⅸ/Ⅴ complex and GPⅠbα of stationary platelet were lower in patients with HTP than those in the healthy subjects. The levels of GP Ⅰ b/Ⅸ/Ⅴ complex and GPⅠbα were recovered after incubation with YQHXD and its part decomposed formulas, but the improving effect were not found in groups of single traditional Chinese drug. Both Astragalus membranaceus and Angelica Scinesis played the most important role in YQHXD, and the expression of receptor of thrombin were enhanced by the compatibility of two herbs. Conclusion The partial mechanisms of treating HTP with YQHXD are attributed to its regulating the expression of GPⅠb/Ⅸ/Ⅴ complex and GPⅠbα, a receptor of thrombin,at the level of receptor protein.
