摘要 目的:体外研究尼古丁对人牙周膜成纤维细胞增殖能力的影响。方法:用不同浓度的尼古丁与体外培养的人牙周膜成纤维细胞作用不同时间,用MTT比色法测定细胞的生长情况,流式细胞分析法测定尼古丁对细胞周期的影响。结果:各浓度的尼古丁组均能抑制人牙周膜成纤维细胞的生长,但不同浓度尼古丁对牙周膜细胞的抑制作用无明显差异;各浓度组尼古丁均能降低G1的比例,高浓度尼古丁(5×10-1g/L)组G1期比例降至84.62%,G2/M期升高至12.83%。结论:尼古丁能抑制人牙周膜成纤维细胞的生长,并影响其细胞周期的进程。 Objective: To investigate the effects of nicotine on the proliferation of human periodontal ligament fibroblasts in vitro. Methods: Human periodontal ligament fibroblasts were cultured in the presence of nicotine at various concentrations. The growth activity was measured with MTT method and cell cycle was tested using flow cytometry. Results: The proliferation of the periodontal ligament fibroblasts was inhibited by the nicotine, but there was no significant difference among different concentrations. The percent of G1 was decreased when nicotine was cocuhured with the cells, the percent of G1 was decreased to 84.62% at high concentration (5 × 10^-1 g/L) of nicotine, the percent of G2/M was increased to 12. 83%. Conclusion: Nicotine inhibits the growth and cell cycle of human periodontal ligament fibroblasts in vitro. It nicotine may have pathological role on human adult periodontitis.
