摘要 β-1,3-葡聚精酶的生产菌种由冻土毛霉经紫外线诱变获得,据正交试验确定培养条件。经硫酸铵盐析和SephadexG-100柱层析,酶得到纯化,并对酶的性质进行了研究。酶对酵母自溶有明显促进作用,使自溶产物的游离氨基氮含量、还原糖含量、总蛋白质得率、干物质得率均得以提高。 A high mutant Mucor hiemalis sabin I- 331 was obtained by using ultraviolet ray.The proper conditions for enzyme production were : 1% yeast, 20% flask's volume of culture,initial pH5.0, culture bine 48h. The enzyme was purified by using ammonium sulfate fractiontion and Sephadex G-100 gel filtration. The optimum conditions for enzyme's action were 50℃and pH6.0. The enzyme was stable over 45-50℃ and pH4.5-7.0. The β-1,3-glucanase hadstrong lytic activity towards the baker's yeast and the brewer's yeast. The effect of autolysiswas improved while a little amount of enzyme (1.3u Per gram of dry yeast) were added in atthe beginning.
