摘要
本文研究乳酪蛋白源抗高血压活性肽(Antihypertensive peptides of casein)的制备及其在体内与体外对高血压的影响。乳源酪蛋白酶解产物经过大孔树脂DA201C脱盐处理,并采用RP-HPLC的分离纯化,检测了脱盐酶解产物与活性肽产物对血管紧张素转化酶(ACE)的抑制活性,并用活性肽产品与酶解物灌胃SHR,观察SHR血压的影响。结果显示,活性肽(0.2 mg·mL^(-1))与酶解产物(1 mg·mL^(-1))对ACE的活性有很强的抑制作用,抑制率分别为75.5%、80.1%;活性肽(5 mg·kg^(-1))与酶解产物(500 mg·kg^(-1))均对自发性高血压大鼠(SHR)有较显著的降血压作用。
Preparation of antihypertensive peptides from casein and its hypertensive effects were investigated in this paper. Hydrolysate of casein was desalinated by macroprous adsorption resins DA201C. Antihypertensive peptides were purificated by RPHPLC. ACE inhibitory activity and antihypertensive activity of hydrolysate and peptides were measured. The results showed that hydrolysate (0.2 mg·mL^-1) and peptides (1 mg·mL^-1) had significant inhibitory activity against ACE (the rate of inhibitory 80.1%, 75.5%), the concentration (the dosages of 5mg·kg^-1 of body weight every day) of antihypertensive peptides had remarkable antihypertension on SHR, and also hydrolysate (the dosages of 500 mg· kg^-1 of body weight every day).
