摘要
白血病多药耐药细胞K562/VCR细胞内Dox浓度明显低于对照组,而P-GP表达则明显高于对照组,提示P-GP的过度表达可能是肿瘤细胞产生MDR的机理之一。CaS及CaMAS可下调K562/VCR P-GP的表达,增加其胞内Dox浓度,表明CaS和CaMAS逆转肿瘤细胞MDR的机理也与降低细胞膜P-GP表达有关。
Meaningfully,the results of this study clearly showed that the doxorubicin level in the multi-drug resistant (MDR)leukemic cells(K562/VCR cell lines)was much lower than that in control group,whereas the P-glycoprotein(P-GP)level was significantly higher than that in control group,suggesting that the excessive expression of P-GP might pity an essential role in the mechanism of producing the MDR nature of tumour cells. Furthermore,the data also demonstrated that CaS and CaMAS could modulate downwards the P-GP expression in the K562/VCR cell lines and increase the doxonibicin level in these cells .thereby indicating that the mechaniam of reversing the MDR tumour cells with CaS and CaMAS could be associated to the decrease of the P-GP expression on cell metnbrance.
