摘要 目的:探讨大鼠死后不同时间次黄嘌呤鸟嘌呤磷酸核糖转移酶(HGPRT)mRNA的降解情况与死亡时间(PMI)的关系。方法:将36只大鼠断颈处死后置于20℃温度控制系统内,利用一步法荧光标记RT-PCR技术检测大鼠脑管家基因HGPRT mRNA在死后即刻至8d的相对表达量的变化。结果:在死后即刻至7d大鼠脑组织均可检测到HGPRT mRNA,其扩增产物呈下降趋势。结论:一步法RT-PCR技术检测大鼠脑组织HGPRT mRNA,在死后的降解呈现出一定的规律性,可为PMI推断提供一种新的指标。 Objective:To explore the relationship between postmortem interval (PMI) and timedependent degradation of Hypoxanthine guanine phosporibosyltransferase(HGPRT) mRNA in the brain of rat. Methods : 36 rats were killed by cervical dislocation and placed in a chamber with temperature-controlling system at 20℃ for 0 to 8d postmortem. Changes of house-keeping HGPRT mRNA in brain were examined by one-step fluorimetric reverse transcriptase-polymerase chain reaction(RT-PCR). Results :The amplification production of HGPRT mRNA could be detected in brain within 7d postmortem, which showed a decreasing tendency. Conclusion:Time-dependent degradation of HGPRT mRNA in brain detected by one-step fluorimetric RT-PCR technique may provide a new indicator for estimation of PMI.
