糜蛋白酶抑制法测定血清ASTm同工酶

Chymotrypsin Inhibition Method for the Determination of AST-m Isoenzyme in Serum

采用人血白蛋白保护ＡＳＴｍ，糜蛋白酶选择性抑制ＡＳＴｃ，采用酶反应速率法测定血清ＡＳＴｍ同工酶的活性。经实验证明：本法抑制系统对ＡＳＴｃ选择性抑制效果达１００％，用３％人血白蛋白加入测定ＡＳＴｍ不受影响，且测定ＡＳＴｍ活性在５００ＩＵ／Ｌ以下呈良好线性；３份高低不同活性的血清标本天内ＣＶ分别为１．４％、４．３％和６．７％，天间ＣＶ分别为１．８％、５．８％和７．７％；３份高低不同活性的血清ＡＳＴｍ平均回收率为９９．６％；与层析法同时测定比较ｒ＝０．９９７５，ｙ＝０．９８５ｘ＋０．２８；临床参考值为７．５ＩＵ／Ｌ以下；能自动化分析。

Serum AST-m isoenzyme with human albumin as the protector and chymotrysin as the selective inhibitor of AST-c,was determined by enzyme rinetil method.The vesults are described as follows: The activity of AST-c was entirely inactivated by the inhibition system.The measurement of AST-m was not affected by 3% human albumin,and the linear range was 0～500IU;Three samples containing AST- m of high and low activity were assayed,The CV within day was 1.4%、4.3% and 6.7% respctively, The CV pacing day was 1.8%、5.8% and 7.7%.The average recovery was 99.6%; The method was compared with chromatography (r=0.9975,y=0.985x+0.28);The clinical reference value was 0～7.5IU;This method also can be used for automatic analysis and applied in clinical practice.
