摘要
为了研究涎腺腺样囊性癌对I型胶原的侵袭能力,本实验利用鼠尾胶原作培养基,接种涎腺腺样囊性癌细胞系ACC2和高转移细胞株ACCM,以研究其侵袭能力。结果发现,肿瘤细胞紧密粘附胶原上,部分形成单层或复层,高转移细胞(ACCM)形成侵袭灶,并形成类似腺管样结构,说明高转移细胞株对I型胶原具有更强的侵袭能力。
Objective: In order to study invasion ability of salivary adenoid cystic carcinoma (Acc) cell on collogen I.Methods: We used Acc-2 cell line derived from a minor salivary Acc, and Acc-M clone, cloned from Acc-2 with high lung metastasis ability. The two cell lines were cultured on the surface of rat tails collager, which composed of collagen I. They were culcubated in 37℃ in RPMI 1640 for two weeks. Then they were observed with light microscope and electron microscope.Results: The result showed carcer cell can grow on the surface, part Acc-M invade into collagen I, and form microcyst-like structure, Acc-M showed a stronger invasion ability than Acc-2.Conclusion: High metastatic clone Acc-M have a stronger ability invading into collagen I. meanwhile it can provide a model of metastastic research.
