摘要
采用PCR-RFLP对奶牛嗜乳脂蛋白基因的第7外显子进行了扩增,并用HaeⅢ进行酶切,产生了2个等位基因A(331+170 bp)和B(295+201 bp)。分析不同基因型对个体乳脂率、乳蛋白、乳糖、干物质和305 d产奶成年当量等生产性能的影响。结果表明:AA基因型和A等位基因的频率分布在所试的4个群体中都较高(>60%),AA基因型奶牛的乳脂率、乳蛋白含量显著高于BB基因型(P<0.05),同时乳固形物含量和乳糖的含量也高于BB基因型,但不存在显著性差异;另外,AA基因型奶牛的305 d产奶成年当量则显著低于BB基因型(P<0.05)。对305 d成年当量、乳脂率、乳蛋白质含量、乳干物质含量和乳糖含量A基因的加性效应分别为-345.34 kg/胎次、0.07%、0.09%、0.09%和0.07%,A等位基因的显性效应分别为187.72 kg/胎次、0.07%、0.08%、0.04%和0.04%。
The polymorphism of exon 7 in butyrophilin (BTN) gene was detected by PCR - RFLP. A Hae Ⅲ RFLP named A (331 + 170 bp) and B (295 +201 bp) was found in the lines tested. Association of the polymorphism with 305 d matured equivalency (ME), fat percentage (F/%), protein percentage (P/%), solids percentage (S/%) and sugar percentage (Su/%) were analyzed. The results showed the distribution of AA genotype and A allele frequency was higher ( over 60% ), and the cows carried AA genotype had higher F% (P 〈 0. 05 ), P% ( P 〈 0.05 ), S% and Su% than those of the cows with BB genotype, respectively, while the ME of cows carried AA genotype was lower than that of the BB genotype ( P 〈 0.05 ). The additive effects of allele A was -345.34 kg/parity,0.07%, 0.09%, 0.09%, 0.07% for ME, F( % ), P( % ), S ( % ) and Su( % ) ,respectively,and the dominant effects of allele A was 187.72 kg/parity,0.07%, 0.08%, 0.04% and 0.04%, respectively.
