摘要
比较分析了I-KI染色法、蓝墨水染色法、TTC染色法、过氧化物酶染色法和离体萌发法5种花粉生活力检测方法在测定中国沙棘花粉生活力中的适用性,并探索了花粉适宜的离体萌发条件.结果表明,I—KI染色法、TTC染色法未能将中国沙棘花粉染上色,因此不适合其生活力的检测;蓝墨水染色法和过氧化物酶染色法能够快速地测定中国沙棘花粉生活力,但测定值均偏高,误差相对较大;离体萌发法适合中国沙棘花粉生活力的测定.在筛选中国沙棘花粉适宜的培养基时,发现中国沙棘花粉在蔗糖(O.15~0.20g·mL^-1)+硼酸(0.1×10^-4g·mL^-1)的培养基上萌发速度较快,有活力的花粉能够完全萌发且萌发率趋于稳定;附加琼脂的固体培养基对花粉的萌发影响不大.
Five pollen viability detection methods, I-KI coloration, blue-ink coloration, TTC coloration, peroxide enzyme coloration and in vitro germination, are adopted to detect the pollen viability of H. rharnnoides L. ssp. sinensis Rousi, and these methods are compared with each other. The suitable conditions of pollen germination in vitro are explored as well. The results show that the methods of I-KI coloration and TTC coloration can not stain the pollen of H. rhamnoides L. ssp. sinensis Rousi, so they are not proper to detect the pollen viability. The blue-ink coloration and peroxide enzyme coloration methods can detect the pollen viability quickly; however, both of the measured value and error of the data are relatively higher. The in vitro germination method is fit to detect the pollen viability. When supplemented with 0.15-0.20 g·mL^-1 sucrose and 0.1 ×10^-4g·mL^-1 Boric acid on the media, the pollen of H. rharnnoides L. ssp. sinensis Rousi germinate faster and the viable pollen can germinate completely. Solid medium supplemented with agar has little influence on the pollen germination.
