摘要
建立了以离子液体为电解质,测定元胡药材中延胡索乙素含量的毛细管电泳新方法。探讨了离子液体浓度、pH值、分离电压对分离效果的影响以及分离的机理。在以石英毛细管为分离柱,50 mmol/L 1-丁基-3-甲基咪唑四氟硼酸盐(pH 7.1)为电泳介质,分离电压20 kV的条件下,延胡索乙素可与其它组分在5m in内完全分离。延胡索乙素的线性范围为31.25-500 mg/L,检出限为4.86 mg/L,回收率为95%-102%。
A capillary electrophoresis method was developed for the quantification of tetrahydropalmatine in the medicinal herbs Rhizoma corydalis by using ionic liquid as electrolyte. The effects of several parameters such as ionic liquid concentration, pH value and applied voltage on the separation efficiency were investigated. The separation mechanism was also discussed. Under the optimal conditions, using pH 7.1 50 mmol/L 1-butyl-3-methylimidazolium tetrafluoroborate ( [ bmim ] BF4 ) as electrolyte, applied voltage of 20 kV and detection wavelength of 254 nm, tetrahydropalmatine was successfully separated fi'om extracts of Rhizoma corydalis in 5 rain. A good linearity was observed in the range of 31.25 -500 mg/L with a detection limit of 4. 86 mg/L. The intra-day RSD was 0. 18% for migration time and 0. 46% for peak area. The inter-day RSD was 2. 65% for migration time and 3.24% for peak area. The recoveries of tetrahydropalmatine ranged from 95% to 102%.
