己酮可可碱对肾缺血-再灌注损伤大鼠的保护作用 被引量:5

摘要 目的探讨己酮可可碱(PTX)预处理对大鼠肾缺血-再灌注(IRI)损伤时细胞间黏附分子-1(ICAM-1)表达的影响,探讨PTX对肾脏的保护作用及其可能的机制。方法SD大鼠75只,随机分为3组:假手术组(SO组)、缺血-再灌注组(IRI组)、PTX治疗组(PTX组),每组25只。通过无损伤动脉夹阻断双肾血流45 min建立大鼠的肾IRI模型,缺血前PTX预处理。3组分别于缺血前、缺血-再灌注即刻、再灌注后1,4,24 h 5个时间点监测肾功能、肾脏病理及肾组织匀浆的ICAM-1、肿瘤坏死因子(TNF)-α水平变化情况。结果PTX组与IRI组相比,肾脏病理改变明显减轻,炎性细胞浸润减少,肾组织匀浆的ICAM-1水平、TNF-α水平显著下降。结论PTX可能通过降低肾组织中ICAM-1、TNF-α表达,抑制炎症、黏附级联反应而减轻肾IRI。 Objective To investigate the effect of penotoxifylline (PTX) on the expression of ICAM-1 in renal ischemia-reperfusion injury , and to evaluate the protective mechanism of PTX on renal. Methods The rat model of renal IRI was established with occlusion of left and right kidney pedicle for 45 minutes. 75 SD rats were randomly divided into three groups: the sham operated controls, IRI group and PTX treated group. The serum creatinine (sCr) values and the levels of ICAM-1 and TNF-α in kidney tissue were measured before operation and 0, 1,4, 24 hours after reperfusion. Results The renal tissue histophathology was ameliorated in PTX-treated group. The valuels of sCr , the scores of renal tubules, the levels of ICAM-1 and TNF-α in kidney tissue in PTX-treated group were significantly lower than those in IRI group. Conclusion PTX pretreatment can attenuate kidney IRI by reducing expression of ICAM-1 and TNF-α in kidney tissue.
