摘要
试验拟建立绵羊精原干细胞移植受体内源性生殖细胞的消除技术。将8只5月龄小尾寒羊公羊随机分为4组,各组第2次处理后3周,公羊睾丸活体采样,石蜡切片、HE染色验证处理效果。结果表明,白消安处理公羊可以完全消除内源性精原干细胞和其他生殖细胞。LHRH、LH疫苗处理的公羊,只能使部分精子发生丧失和生殖细胞消除。白消安是精原干细胞移植受体绵羊内源性生殖细胞最有效的消除技术。
In order to establish the technique of depleting endogenous germ cells for transplantation recipients of spermatogonial stem cells in ovine, eight of 5-month-old small-tail Han rams were randomly divided into four experimental groups to test. After three weeks from the second treatment, the samples of testes are obtained in vivo. The testes samples were embedded in paraffin, section cut, stained with hematoxylin and eosin, and observed under a microscope. The results show that the endogenous spermatogonial stem ceils and other germ ceUs can be depleted absolutely by treatment of busulfan. There are part of sperm lost and germ cells depleted by treatment of LHRH and LH bacterin. The chemotherapy with busulfan is the most effective technique to deplete endogenous germ cells for transplantation recipients of spermatogonial stem cells in ovine.
